SNP-based assessment of genetic purity and diversity in maize hybrid breeding

Assessment of genetic purity parental inbred lines and their resultant F 1 hybrids is an essential quality control check in maize hybrid breeding, variety release seed production. In this study, purity, parent-offspring relationship diversity among the were assessed using 92 single-nucleotide polymorphism (SNP) markers. A total 188 genotypes, comprising 26 lines, four doubled haploid (DH) 158 single-cross investigated study Kompetitive Allele Specific Polymerase Chain Reaction (KASP) genotyping assays. The bi-allelic data was analyzed for parameters GenAlex software. SNP markers highly polymorphic 90% had information content (PIC) values > 0.3. Pairwise distances ranged from 0.05 to 0.56, indicating a high level dissimilarity lines. maximum distance (0.56) observed between CKDHL0089 CML443 while lowest (0.05) I-42 I-40. majority (67%) studied genetically pure with residual heterozygosity <5%, only 33% levels >5%. Inbred which not pure, require purification through further inbreeding. Cluster analysis partitioned into three distinct clusters potential contribute new beneficial alleles breeding program. Out 68 (43%) that passed test, seven namely; SCHP29, SCHP95, SCHP94, SCHP134, SCHP44, SCHP114 SCHP126, selected as candidates evaluation due outstanding yield performance.
